Kristen Wiig, and Seth Rogen are casting directors looking for actors and actresses for an upcoming movie about a man and woman who decide to say they are in love with their cousins so they can get away with being single well into their 30's. The set up is 7 auditoriums filled with people going out for the role, with Kristen, Seth, and a different casting director for each room sitting at a table with microphones, as they look for the perfect star to fill their coffee mugs. We see Kristen, who is bubbly and full of joy at work, intercut with scenes of her at home: miserable, alone, and looking for someone to love. Seth, who is sarcastic and egotistical at work, is shown at home as an insecure, self reliant man on a mission to find his long lost sister, who came to LA to be an actress.

The 7 auditoriums include real actresses and actors of Hollywood, mixed in with people from LA looking for their big break. Auditorium 1 has blonde haired women only. Auditorium 2 has brunette haired men. Auditorium 3 has red haired children with their parents in a room with a window. Auditorium 4 has African American men and women together in one room. Auditorium 5 has red haired women with freckles, and dressed in their favorite movie character. Auditorium 6 has Black haired men who are told to prepare a monologue that requires a prop. Auditorium 7 has blonde haired men who are told to memorize the same line, and be able to cry on cue.

Bill Hader, Dave Franco, Seth MacFarlane, Julia Louis Dreyfus, Chris Kattan, Jennifer Aniston, Fred Armisen play the third casting director in the auditorium.

They start to divide people into "non Special" and "O" groups in each room.
They send everyone out into LA for a "live" audition where they must complete a task as a character that doesn't know what money, stop signs, or coffee is.
They have people stand in circles and tell each other out loud what their favorite planet is, and why.
They allow actors and actresses to leave the room - before auditions roll - if they think the role is beneath them. Meryl Streep is the only one to leave, as she forgets her purse.
They do one on one casting, where Kristen starts to have feelings for certain actors because of the scene they read together. She starts to favor people that Seth Rogen hates.
They have people wear makeup that makes them look like hookers, and many actresses and actors start to question the production of the film, and have their agents show up with them.
The director stops by with a script that is red, while everyone in the last round is given scripts that are brown, and the story starts to focus on these 3 actresses who decide to get their hands on the red script to see if they can sell it.
And Julia Louis Dreyfus decides to sue one of the actors for sexual harassment after one of the blonde haired men says his line with a wink and a smile, and this reminds Julia of her ex husband, and she starts to question whether she should sue the studio.
